help, I'm not sure if the tube I made for the queen ant will fit

I place the queen in a straw with dry and wet paper towels on either side. In order to prevent water loss, I put a plastic bag on the wet paper towel and tied it tightly. Is this condition enough for the queen to reproduce?

I do not think that will hold water in well. It may slow down evaporation, but you will want something more like a test tube setup if you do not want to replace the water regularly.

I would get a proper test tube setup if you can. They’re easy to find in bulk on Amazon for cheap.
