So I got this camponotus queen in this enclosure I got from tarheel ants and I am using it as a way to found a colony as the queen kept digging out of her test tube, but would this work if I kept it somewhere dark and undisturbed?

I have only ever successfully cultivated a colony in a test tube setup. I'm assuming that drop of liquid on the floor is honey? I would advise removing honey because over time it will become more sticky and the queen can become entrapped. The queens can go without eating for quite some time so there is no need for food.

It should as long as you keep enough moisture in it. Maybe some wet cotton? But if you can find some way to keep her inside the test tube while still allowing proper airflow go with the test tube, as it is a 10x better way to found a colony in my opinion.
