So I stopped by the pond today and I lifted up this rock and hoped that there were ants to check out. I had a test tube ready prior to walking down there and I got some ants. Being so interested into ants I put them in my homemade Formicarium and watched them. Then I noticed that one of the ants (black) was silghty lager than the other ones (workers) it's nodes were stretched out further and its gaster was bigger and had 2 stripes. Is this a queen ant?

To answer you question, this is definitely not a Lasius niger (I don't mean to be rude, but this is the correct spelling.) queen from what you have described.

We wouldn't know what kind of ant you were actually looking at to determine if one was a queen or not. Workers that are full of food may have much larger gasters than their sisters. If you can take pictures and let us know where in the country you found them, maybe we can figure it out.

If you've enjoyed using my expertise and identifications, please do not create undue ecological risk by releasing your ants. The environment which we keep our pet insects is alien and oftentimes unsanitary, so ensure that wild populations stay safe by giving your ants the best care you can manage for the rest of their lives, as we must do with any other pet.
Exotic ants are for those who think that vibrant diversity is something you need to pay money to see. It is illegal to transport live ants across state lines.
