I have a 5 queen colony of Tapinoma that have a few workers and a lot of brood. I have found a lot of workers outside near my house. I have read that the colonies are very interconnected. Would I be able to add a few workers from outside to the current colony in the test tube?

Probably. But I never have kept them, so wait for everyone else to answer before acting.
My Ants:
(x1) Campontous semitstaceus ~20 workers, 1 Queen
(x1) Camponotus vicinus ~10 workers, 1 Queen (all black variety)
(x1) Tetramorium immigrans ~100 workers, 1 Queen
(x1) Myrmercocystus mexicanus -1 Queen
(x2) Mymercocystus mimcus -1 Queen
(x1) Mymercocystus testaceus ~45 workers, 1 Queen

You can, but I don't see that point. It takes away a lot of the fulfillment and joy antkeeping gives and can even be somewhat harmful to the colony long term. T sessile are such a fast growing species, even with one queen (and you have 5!) that the difference will be minimal and will quickly get covered up by their biological daughters.

It was Brachymyrmex actually...


Ants I have: Tapinoma sessile(2 queen colony). RED MORPH Camponotus neacticus(now has pupae!), Tetramorium immigrans (x3), Aphaenogaster sp, Temnothorax sp, Brachymyrmex sp.   possibly infertile   :(,  Ponera pennsylvanica, and Pheidole morrisi!  :yahoo: 

 

Other insects: Polistes sp. Queen

                    

Ants I need: Pheidole sp., Trachymyrmex sp., Crematogaster cerasi , Dorymyrmex sp. Most wanted: Pheidole morrisii
