How do you tell founding chambers from mature colonies or other insects such as termites? I often come across holes but don't dig because I can't tell the difference. Thanks

Look for the queen bringing dirt out or watch to see if workers are coming out. Between finishing the chamber and getting workers, I don't know. Maybe watch it for awhile and if you don't see anything give it a shot? I just don't dig for anything.
Real nests are big. Founding nests are this tiny hole. I just dig whatever small hole I see. If I were you I'd probably do the same because if it was an actual colony the hole would be deeper so it wouldn't matter.
I have found founding queens while doing landscaping projects.
Go to the ant, you sluggard; consider its ways and be wise! It has no commander, no overseer or ruler, yet it stores its provisions in summer and gathers its food at harvest. -Proverbs 6: 6-8
