If I connect to my test tube a formicarium with sand is it a problem for them? The queen can lay eggs on top of the sand? And I need to boil the sand?

What species are your ants, and what size is your colony? Are they supposed to dig tunnels in the sand? If so, I would recommend using coconut husk which can be found at many garden centers, instead. It absorbs water, holds its form better, and is probably closer to the ants' natural habitat depending on where you live. If possible, avoid digging medium altogether (Unless you prefer it) and look for tutorials on Formiculture instructing you on how to create your own nest.

If I got a test tube with a queen and some eggs and larvae but none workers I don't need to feed her until the workers will hatch right?

Depends on the species. However, if the queen already has larvae, then most likely yes.
