If I got a test tube with one queen and some larvae and eggs but no workers I don't need to feed her until the workers hatch right?

It would depend on what species of ant she is. Most species are claustral which means they hole up and wait until their brood are ready to bring them food, they generally never leave their nest. Some are semi-claustral which means they will leave their nest to scavenge for food even absent of brood. It would be beneficial to know what queen/species you are working with.

So then the answer is yes, you may leave them alone and do not need to feed them. Check back in a month!
