I have plenty of left-over plexiglass and I want to see if I can build a nest out of them.

You can do the three stack method. Two on the outside, and the one in the middle is cut in the desired shape of the tunnels and chambers inside.

Could I, for larger ants, make 4 stacks?

Of course. Plexiglass (acrylic) can be easily bonded together by welding it using a solvent. Look up "acrylic welding". There are all sorts of tutorial videos on Youtube.

I made one from acrylic.

It didn't work too badly, but the ants definitely enjoyed it more when I put some grout on the floor chambers.  I heat mine so condensation is a big issue, and the grout solves most of the problem.
